The forms of SNAC:
We usually refer to the abbreviation SNAC as SNAC powder. In addition to the powder state, there are SNAC crystal form, methanol-water cosolvent compound of SNAC and ethanol-water cosolvent compound of SNAC.
SNAC powder, also known as amorphous SNAC, is unstable when exposed to environmental conditions and changes to the SNAC crystal form when exposed to ambient moisture. In this case, the crystal form is SNAC crystal III.
There are six forms of SNAC crystal, details as follows:
1.SNAC crystal I. SNAC crystal I is anhydrous crystal form, stable at room temperature, and does not change crystal shape when subjected to grinding or pressing. However, it can be transformed into SANC crystal III by wet granulation under appropriate conditions.
2.SNAC crystal II. SNAC crystal II is the hydrate of SNAC, specifically 1/3 hydrate (SNAC 1/3 hydrate refers to the crystal of SNAC, in which one molecule of water is combined with three molecules of SNAC). Crystal type II is stable at room temperature. Non-crystal II SNAC can be converted to crystal II using drying or contact methods.
3.SNAC crystal III. SNAC crystal III is a hydrate of SNAC, specifically a trihydrate (SNAC trihydrate refers to the crystal form of SNAC, in which three molecules of water bind to each molecule of SNAC), and crystal III is stable at room temperature. Other crystal forms of SNAC can also be made to crystal III by wet or dry process.
4.SNAC crystal IV. SNAC crystal IV is anhydrous. Crystal type IV is stable at room temperature. Other crystals of SNAC can also be made into crystal IV by heating.
5.SNAC crystal V. SNAC crystal V is a methanol-water cosolvent compound (SNAC: methanol: water is 1:0.8:2). Other crystal forms of SNAC can also be converted to crystal V of SNAC in corresponding ways.
6.SNAC crystal VI. SNAC crystal VI is an ethanol-water co-solvent compound (SNAC: ethanol: water is 1:0.6:2). Other crystalline forms of SNAC can be crystallized in ethanol solution to prepare crystal VI.
Above, the powder state and crystal type I-IV of SNAC can coexist at room temperature without affecting its using effects.

The following is a preparation method of SNAC crystal I or III and heparin USP capsules:
1.Weigh heparin and SNAC and transfer to a clean, dry 8oz glass cream bowl, SNAC crystal I 153.33mg or SNAC crystal III 181.72mg and heparin USP 56.82mg.
2.Add heparin equivalent volume of SNAC to the cream bowl and mix with pestle for 2 minutes.
3.Add the rest of the SNAC to the mixture and mix for another 2 minutes.
4.Fill with capsules.
