The benefits of SNAC  

SNAC, short for Salcaprozate sodium, is a major intestinal osmotic enhancer (PE) that can accompany poorly permeable macromolecules across the plasma membrane in a transcellular mode of action.  Thus, it is possible to increase oral absorption of several poorly permeable molecules that are jointly administered.  

Up to now, SNAC is the most extensively tested vector and the only one approved PE in oral formulations.

In one study, six study participants were recruited and tested for ingestion of MK677 powder alone and MK677 powder containing SNAC.  The study dissolves MK677 powder and SNAC in ethanol aqueous solution (because ethanol aqueous solution can effectively dissolve MK677 powder and SNAC).  

One group mixed MK677 powder with 15ml(about one tablespoon) of aqueous ethanol, and the other group mixed MK677 powder with aqueous ethanol and a premixed solution of 200mg SNAC.  

Comparative experiments show that:

1.For the test subjects who ingested MK677 powder containing SNAC, the effect was produced within 5 minutes, and the time limit for the initial effect was between 2 minutes and 5 minutes;  For the test subjects who ingested MK677 powder without SNAC, the time to effect was 15 minutes, ranging from 15 minutes to 75 minutes.  

2.For the test subjects ingesting MK677 powder containing SNAC, the effect reached a peak of 4.7 30 minutes after ingesting the powder;  For those who ingested MK677 powder without SNAC, the effect peaked at 2.2 after 135 minutes.

3.The results showed that the addition of SNAC did not shorten the action time of MK677 powder.  

4.The above experiments indicate that the addition of SNAC to oral preparations can provide a faster time of action and a higher intensity of action at peak activity levels.  Moreover, the addition of SNAC had no effect on the duration of action of the product.
